The multi-beam SIM method does not only improve the resolution of SIM images by three times, it also reduces the required image acquisition time. The objective lens is redesigned to optimize the signal as expected for multi-beam SIM with an NA of 1.33. The lateral resolution increases from 15 nm to 9.5 nm and the axial resolution increases from 9.4 nm to 5.9 nm (see Fig. 3)24. While the spatial resolution obtained by regular SIM is limited by the incident light illumination (which is diffraction-limited), the diffraction limit is significantly reduced in the direction orthogonal to the illumination. When this is taken into account, the lateral resolution improves by a factor of approximately 3 (axial: factor of 10) and the image acquisition speed is improved by a factor of approximately 6 (time-lapse: factor of 3). Multi-beam SIM images can be reconstructed after only three images with the resulting lateral resolution of 6.7 nm and axial resolution of 3.4 nm (see Fig. 3 and Supplementary Movie 3).

However, two-color SIM imaging is possible and provides additional contrast due to the shift of fluorescence emission into the different SIM channels20,22,23. The performance of the system is significantly affected by the thickness of the biological sample; with maximum possible thickness of 100 µm, the SIM accuracy degrades to 50 nm and the time-lapse imaging rate drops to 30 Hz (see Fig. 2, Supplementary Fig. 4 and Supplementary Movie 2).
